| Research Abstract |
1. 平成10年度は、中国雲南省、遼寧省および新疆省地方に於いて新しく採集された植物は112種であり、これらの植物はアルコールおよび熱水により抽出された。中国において凍結乾燥機の不調のため、現在アルコール粗抽出物112種が調製されている。
これらの粗抽出物について、がん培養細胞毒性など数種の評価系について検索が進行中である。昨年までに1次スクリーニングされた粗抽出物についても有効物質単離精製のために多量の植物が採取された。
2. Polygonum cuspidatum Sieb.et Zucc.(虎杖)の根の粗抽出物よりP388/ADMおよびK562/ADM培養細胞に強い細胞毒性を示す2種の物質が単離され、機器分析の結果、physionとemodinと同定された。後者はより強い細胞毒性を示した。
3. Carthamus tinctorius(紅花)の熱水抽出物よりマウス脾細胞幼若化促進物質SFlとFS2が得られた。SFlはマウス腹腔内滲出細胞によるIL-1、IL-6およびTNF等の強いサイトカイン産生誘導活性を示した。またマウス移植FM3A腹水癌に対する抗がん活性およびB16メラノーマ細胞の肺転移抑制作用が認められた。
4. Wikstroemia indica C.A.Mer(了哥王)およびCaesalpinia sappan L(蘇木)の粗抽出物はtsFT210細胞を用いたフローサイトメトリー法による検索で細胞周期のG2/M期を強く阻害した。阻害活性物質の単離精製および化学構造について追究中である。
5. Canarium bengalensis Roxb(方欖)の粗抽出物より、2種の抗酸化活性物質が単離された。1つは1,4-dihydroxybenzeneと同定された。もう1種の化学構造は追究中である。
6. Plectranthus excisus Maxium(尾叶香茶菜)の粗抽出物より、エピネフリン存在下ラット脂肪細胞から誘導される脂肪酸の遊離を阻害する物質reniformin AおよびKamebakaurinを単離同定した。これら活性物質の作用機構について追究中である。
