Research Abstract

本研究は、BCP前駆体の立体構造および活性化機構を明らかにする目的で実施されている.本年度実績の概容は以下のとうり.
1: 大量のBCP前駆体がたの精製に成功する(200mg)
2: 高圧電子顕微鏡による蛋白質のネガティブ染色像を得ることに成功する(現在継続中)
3: CDスペクトルを用いた活性化の際の立体構造の変化についてデータを得て現在コンピューターによりデータ解析中.
4: In vitro mutagenesis法を用い、プロペプチドの活性阻害部位を検討した.
5: X-線解析用の予備的結晶をえた.
6: これらの過程で山本は、これまでに知られていなかった全く新たなシステインブロテアーゼの阻害蛋白質(BCP特異的)を発見しその精製、cDNAのクローニングに成功した.アミノ酸および塩基配列の両面から全一次構造を決定した.
