| Research Abstract |
【緒言】本研究では、独自に開発した遺伝子解析法を用いて、個々の癌および末梢血などの非侵襲的検体の遺伝子異常を広範に検索し、癌化の機構解明と新しい癌の遺伝子診断法や早期発見法の確立を目的とした。
【方法】1,マクロな遺伝子異常検出法としてInter-Alu Long-PCRゲノムスキャン法を,2,ミクロな遺伝子異常検出法として遺伝子全領域マルチSSCP法と蛍光標識マイクロサテライト(MS)解析法を開発した。
【結果】1の方法によりヒト腎癌における新しいヘテロ接合性消失(LOH)領域を発見した。RH法とMS解析によりこの欠失領域をD14S1005付近に限定し、更にこの領域のBACクローンを分離して、2種の癌抑制遺伝子候補を同定して解析している。同じMS解析法により、頭頚部癌がヒト染色体13q33領域で高頻度のLOHを示すことを見い出し、この領域に局在する癌抑制遺伝子候補、ING1遺伝子の全体構造とプロモーター領域を解明し、3例の癌で癌特異的な不活化点突然変異を初めて発見した。更に、MS解析によって染色体7q31領域に各種の癌で高頻度のLOHがあることを確認し、この領域に存在するING3遺伝子の発現が頭頸部癌の多くで減少していることを発見した。その他にも、肺癌、骨髄性白血病、食道癌、肝癌、脳腫瘍などにおける染色体LOH領域を多数確認し、それらの領域における癌抑制遺伝子の候補を計8種同定し、詳細に検討している。以上のほか、骨軟部腫瘍の遺伝子診断を融合遺伝子のRT-PCRによる検出で系統的に行なう方法を確立し、病理診断で不確定であった4例の症例を確定診断することができた。末梢血診断に関しては、血中遊離DNAの超微量定量法を開発して癌の進展や寛解によって変動することを確認し、また血中遊離DNAを用いたras遺伝子群の活性化点突然変異の高感度検出法とp53遺伝子の変異検索の系を確立した。
