| Research Abstract |
本研究において申請者らは、1)抗リン酸化ペプチト抗体群を用いることで細胞質分裂溝の近傍に活性化Rhoキナーゼが集積し、中間径フィラメント蛋白質をリン酸化することを明らかにした。2)また、このRhoキナーゼによるフィラメント蛋白質のリン酸化部位にリン酸化をうけないような変異を導入した中間径フィラメント蛋白質によって構築されたフィラメントは、細胞質分裂が終了しているにもかかわらず、フィラメントの断裂・分配が生じていないことを明らかにした。3)Two hybrid法を用いて、新規中間径フィラメント総合蛋白質を数多く見出した。それらのうちMrj(Hsp40のファミリー蛋白質)の解析は進みMrjが単層上皮に特異的に発現するケラチン8/18中間径フィラメントを選択的にその構築を調節することを明らかにした。
