Research Abstract	エキノコックス症(多包虫症、単包虫症)、有鉤嚢虫症に関する鑑別血清診断法の確立を目指す研究を展開し、それぞれの疾患に特異的な抗原を検出、精製することによって鑑別診断が可能な段階に到達した。多包虫症と単包虫症とについては、それぞれEm18、Antigen Bと呼ばれる抗原を診断指標とするイムノブロット法が有用である。一方、嚢虫症については等電点電気泳動によって部分精製した嚢虫液抗原(Glycoproteins)を用いるイムノブロット法とELISA法とを確立した。嚢虫抗原については中尾がモノクロナル抗体を作製しており、モノクロナル抗体を用いるサンドイッチELISA法の開発を現在検討している。遺伝子診断法についてはミトコンドリアDNAのCO-1遺伝子の比較解析から多包虫、単包虫、嚢虫それぞれに特徴的な塩基配列の解読が進んでいる。種特黒的なブライマーの作製により種の同定、strainの同定が今後容易になると期待される。中尾が中心になって、これまでの一部の遺伝子解析にとどまらず、多包虫ミトコシドリアDNAの全塩基配列の解読に成功しており、この最新の成績を加味することによって来年度の遺伝子解析の研究は大きく進展すると期待される。エキノコックス症についての血清診断法の今後の展開としては、まだ作製に成功してないEml8、Antigen Bsubunit(8 kDa)それぞれに対するモノクロナル抗体作製を計画している。既に手元にあるGlycoproteinsに対するモノクロナル抗体を含め、これらの抗体をプローブとして遺伝子組換え抗原作製へと研究を展開していきたい。
