Research Abstract	身長発育は、極めて遺伝的に規定されているprocessであるが、遺伝子多型と身長の規定に関する研究は少ない。ビタミンD受容体のexon2多型(AT/CG)多型はアミノ酸配列の長さに差が生じ、ACG群がビタミンD受容体DNA結合部位に当たる部分のアミノ酸が3個短いビタミンD受容体をコードすると考えられる。遺伝子変異(Exon2多型)と骨密度・身長・の関媒について検討した。さらに成長板軟骨に発現している遺伝子群で軟骨分化に関係すると考えられる、成長板軟骨移行帯に発現するPTHrPならびにその受容体であるPTH/PTHrP受容体についても遺伝子多型・後者についてはその機能を検討した。 18-20歳の健康女性において、Exon2多型ATG/ACG群の身長が他の2群と比し有意に身長が高かった。この傾向は思春期においても認められた。RT/PCR法ではATG alleleの発現がACGに比し2-3倍であった。PTH/PTHrP受容体については腎臓ならびに骨に発現されるP3promoterのA rich領域にAAAGの反復配列(通常は5個であるが6-7個となる)の多い例を認めた。この過剰配列はLuciferse法でプロモーター活性を滅弱する事を認めた。さらに正確に下肢長を測定し,上節下節比で表きれるプロ歩ーションを示す本邦正常値がないため現在正常値を設定中である。
