| Research Abstract |
細胞外マトリックスは結合組織・支持組織の主要成分としての働きの他に、細胞を取り巻く環境として細胞の形態、発生、分化を初めとした様々な細胞代謝の調節因子でもある。プロテオグライカンは細胞外マトリックス中でコラーゲンと共に主要な構成成分として重要な働きをもっている。本年度は、細胞外マトリックス分解酵素のひとつであるヘパラネースを昨年度確立した精製法を用いて大量調製しその酵素学的性質をさらに詳細に解析した。また、ヘパラネースの分子クローニングから得られた遺伝子情報をもとに、細胞外マトリックス代謝過程におけるヘパラネースの役割を分析した。本年度得られた結果の要約は以下のとおりである。
1.ヘパラネースの基質特異性を昨年度に引き続き解析し切断部の糖鎖構造を解析した結果、この酵素はendo-β-glucuronidaseの一種であり、切断部から非還元末端側の糖鎖配列は-GlcUA-GlcNAc(or GlcNSO_3)-GlcUA-GlcNAc(±SO_3)-GlcUA-(切断端)であることが決定された。
2.モノクロナル抗体、ポリクロナル抗体の調製を引き続き行い、抗体の性質をウエスタンブロット法、組織染色法などを用いて精査中である。
3.ヘパラネースとGFPタンパクの融合タンパクをコードする発現ベクターを設計し、ヘパラネースの分泌と、細胞内局在に関する検索を行っている。
4.肺癌、食道癌、口腔扁平上皮癌の組織を用い、細胞外マトリックス分解過程、癌転移巣形成過程とがん細胞でのヘパラネース発現の間に相関性のあることを見い出し、そのメカニズムについて解析中である。
