| Research Abstract |
マウスにおいて、Fasを介するアポトーシス誘導の分子機構を正確に解析するために、Fas刺激の際に形成される少なくともFas、FADD、caspase-8から成るDISC(death-inducing signaling complex)を同定する必要がある。そのために、マウスcaspase-8を特異的に認識するモノクローナル抗体を作成した(以前から報告されている抗マウスcaspsae-8モノクローナル抗体は非特異的なものであることも示した)。そして、マウスにおけるDISCの解析を可能とし、以前に報告されているマウスDISCが必ずしも正しくないことを示した。また、B細胞において抗原刺激によってFasを介するアポトーシスが抑制される系を確立した。
TRAF6欠損線維芽細胞にTRAF6変異体を適当量発現させ、TRAF6の生理的な機能領域を同定した。その結果、N端側にあるRING finger及び5つの中の最初のZinc fingerはNFκB活性化に必須でなかった。しかし、この領域はMAPK活性化に関与した。またNFκB活性化においてLPSの場合2と3番目のZinc fingerを必要としたが、IL-1は4と5番目を必要とした。本結果は、TRAF6がZn結合領域中に複数の機能部位を有し、その部位により複数の下流タンパク質にシグナルを伝達していることを強く示唆する。
