Research Abstract	本年度は、抗腫瘍免疫誘導に重要な樹状細胞(DC)サブセットの同定、並びにその作用機序について検討した。マウス骨髄細胞をGM-CSF+IL-3の条件下で誘導したDCをBMDC0,GM-CSF+IL-3+Th1サイトカイン(IL-12+IFN-γ)の条件で誘導したものをBMDC1,GM-CSF+IL-3+Th2サイトカイン(IL-4)の条件で誘導したものをBMDC2として実験を行った。その結果、ナイーブTh細胞をBMDC1と抗原の存在下で培養することによってIFN-γを高産生するTh1が誘導され、BMDC2と抗原の存在下で培養することによって、IL-4を産生するTh2が誘導されることが明確にされた。さらに、BMDC1とBMDC2においては、CTL誘導におけるAPC機能においても大きな機能的違いが存在した。すなわちBMDC1は効果的にアロCTL、を誘導することができたが、BMDC2の刺激では強いCTLを誘導することはできなかった。Th1サイトカインで活性化誘導されたBMDC1は、Th細胞の機能をバイパスして、単離したCD8^+T細胞からもCTLの誘導を可能にした。このBMDC1に依存的なCTLの誘導においては、IFN-γの産生は必ずしも必要とせず、むしろIFN-βが重要な役割を果たしていることも明確にされた。また、IL-2やLFA-1依存的なDC/CD8^+T相互作用がCTLにおいては重要なことも明確にされた。現在、このようなCTLやTh1を効果的に誘導させることができるBMDC1サブセットを用いた癌治療への応用をさらに検討している。
