Research Abstract	今年度は以下の研究成果を得た。 1)膵癌PUNではHLA-A26に提示される癌抗原の候補遺伝子をとることに成功した。 2)骨肉腫OS2000ではHLA-B55に提示される抗原遺伝子の侯補を得た。 3)IAPであるサバイビン蛋白由来HLA-A24拘束性2Bペプチド、リビン由来L7ペプチドを同定した。テトラマー解析により2Bペプチド反応性CTL前駆細胞数は多くの患者で上昇していた。他方、L7は高いCTL活性を誘導することが示され、リビン発現腫瘍には癌ワクチンとして優れている可能性が示された。 4)リカバリン由来HLA-A24拘束性R49.2ペプチドはテトラマー解析により多くの癌患者でCTL前駆細胞数が上昇していた。また臨床的な進行癌と相関することも示唆した。 5)滑膜肉腫転座産物由来HLA-A24拘束性SYT-SSXペプチドもテトラマー解析の結果CTL前駆細胞数が滑膜肉腫患者で上昇し、特に肺転移を示す患者にはより著明な上昇が示された。 6)抗原ペプチドの細胞内提示機構について解析した。チロシナーゼ由来ペプチド等、約10種類のペプチド(9-10アミノ酸よりなる)につき、分子シャペロンhsc73結合性を小胞体内への転送効率につき相関性をみた。その結果、hsc73への親和性と小胞体内への転送には正の相関が認められた。
