| Research Abstract |
本研究の第1の目的は我々が独自に有する自家癌-CTL(cytotoxic T lymphocytes)のペアを利用した癌抗原遺伝子クローニングである。今年度は、ヒト自家骨肉腫OS2000でHLA-A24およびB55が提示する腫瘍抗原遺伝子PBFのクローニングが成功した。遺伝子本体はPBF(papilloma binding factor)であり、新しい転写制御因子で、腫瘍に選択的に発現し、新しい腫瘍抗原として大きな期待がもたれる。本研究の第2の目的はreverse-immunological approachで癌で選択的に発現増加する分子あるいは新規に発現する分子を標的としてCTLの応答性を解析することにある。その結果、1)inhibitor of apoptosis protein (IAP)であるsurvivinが腫瘍で選択的に発現増量し、かつこの分子由来のペプチドがHLA-A24拘束性に癌免疫の標的になることを明らかにした。また2)滑膜肉腫にみる染色体転座(X;18)部位由来SYT-SSXペプチドも同様な標的になることも明らかにした。今年度はこれら2つのペプチドの第1相臨床試験をsurvivnペプチドについては大腸癌患者と乳癌患者で、またSYT-SSXペプチドについては滑膜肉腫患者で開始した。
