真菌症の遺伝子診断

Research Abstract

分子生物学的診断法を確立するためまず各菌種のchitin synthase(CHS)遺伝子の塩基配列を解析し、塩基配列情報をGene Bankに登録し、データーベースを構築した。さらに臨床分離株のCHS遺伝子を解析し、作成したデーターベースと比較を行い、分子生物学的手法による菌種の迅速同定法を確立した。作成したデーターベースや同定法については、学術学会やいくつかの国際的な学術雑誌に発表を行った。
次に真菌症の病変部位からの生検材料、血液、排泄物から抽出した試料のDNAからCHS遺伝子のクロニングを行い、塩基配列を解析した。それと作成したデータベースに登録した遺伝子が一致する菌種を検索して、起因菌の同定を行った。この結果と実際に試料から分離した菌の同定結果と比較を行い、我々が作成した遺伝子診断結果と従来の診断結果が一致することを確認した。以上の結果から、我々が確立した遺伝子診断は、従来の手法と比較して、迅速に診断が行うことが可能で、特異性および感度も優れていることが確認された。これらのことについては、すでに学術学会やいくつかの国際的な学術雑誌に発表を行っている。
