Research Abstract	本研究の主軸のひとつであるヒト肝細胞を用いる研究については本学倫理委員会の承認(平成13年)を得て、その後研究に傾注してきた。すなわち、肝炎ウイルス感染を認めず、切除側肝非腫瘍部肝組織(いわゆる正常肝)より肝組織を採取、そして肝組織片に対し可能な限りコラゲナーゼ灌流法を適用し、肝細胞を分離することに始まった。困難を伴っていた第一段階の研究手法が平成14年度においては、成功することができラットにおいて安定的に研究されている小型肝細胞と同様の性格を有する肝細胞で構成されるされていることについても確認しえ、コロニー形成も確認された。すなわち、増殖能力、肝特異的酵素・蛋白の発現・分泌、そして分化については肝実質細胞と胆管上皮細胞の2方向の成熟型細胞への分化を同定しえた。またさらに、細胞移植を目指す前法としての人工肝組織構築においては、コラーゲンスポンジ内での肝組織形成そしてそこでの細胞分化を検討したところ、類肝組織形成とそこでの肝細胞分化についてもその成果を公表できるに至っている。達成し得なかった研究としては、in vivo研究としての無アルブミンラットを用いた治療研究である。一方、ラット小型肝細胞を用いた細胞移植研究については、(1)小型肝細胞への遺伝子導入、(2)遺伝子導入細胞のラット肝内への細胞移植、について知見を得ることができた。追加研究の上、公表できる段階に入っている。平成14年度に行うべき研究についてはかなり達成しえたと思われる。限界点を残した内容については、次研究へと繋ぐことのできる条件を設定することができ、これまでの成果については意義が大きかったと考えられる。
