Research Abstract |
我々は、慢性荷重刺激であるクライミング運動が、1)骨髄細胞において、破骨細胞分化を抑制し、骨芽細胞分化を促進すること、2)骨吸収を早期に抑制し、骨形成率を増加させることによって、脛骨海綿骨量を増加させること、をマウスにおいて明らかにしてきた。 今年度は、非荷重後の再荷重による骨量回復過程において、骨の細胞増殖及び分化調節に関わる遺伝子発現の変化とcyclo-oxygenase-2(Cox-2)の関与の有無を明らかにする目的で実験を行った。8週齢のC57BL/6J、雄マウスを用い、1週間の尾部懸垂を行ったのち再荷重した。再荷重の3時間前に、Cox-2 selective inhibitor (Celecoxib 60mg/kg)を投与する群(Cox群)とvehicleを投与ずる群(V群)を作成した。V群では、1週間の非荷重後2週間再荷重することで、海綿骨量は正常荷重群と同レベルまで完全に回復した。しかし、Cox群では、海綿骨量の回復は部分的であった。再荷重後24時間のマウスから採取した大腿骨と脛骨の骨髄細胞を培養し、アルカリフォスファターゼ陽性CFU-fの形成をみると、V群では正常荷重群と同レベルまで完全に回復したが、Cox群では依然低下していた。次に、再荷重後1、24、48時間の骨髄細胞における遺伝子発現をquantitative RT-PCRで解析した。Cox群では、V群に比べて、c-fos(1、24、48時間の時点で)、osterix(24時間)、IL-11(24時間)が低下していた。cbfa-1、BMP2、osteocalcinは両群間で差はなかった。 非荷重後の再荷重による骨量回復は、Cox-2 selective inhibitorにより部分的に阻害された。この骨量回復過程には、Cox-2依存性と非依存性のシグナル伝達分子があることが示唆された。
