Research Abstract	1.コバラミン酵素の科学と進化工学的改変ジオールデヒドラターゼはグリセロールにより自殺不活性化を受ける。合理的デザインによりグリセロールによる不活性化を受けにくい酵素に改変することを目指す第1歩として、グリセロール結合型ジオールデヒドラターゼ・シアノコバラミン複合体の結晶構造解析を行い、分離能2.13Åまで精密化を進めた。プロパンジオール結合型とグリセロール結合型で、全体及び各サブユニット毎の構造上の違いは見られなかったが、基質の3位水酸基と水素結合しうる2つのアミノ酸残基を特定できたので、今後は変異型酵素の作成を行う。ジオールデヒドラターゼ・アデニニルペンチルコバラミン複合体を暗所および光照射条件で結晶構造解析を行った。可視光照射により補酵素アナログのCo-C結合が開裂し、生じた上方配位子由来のアデニニルペンチルラジカルはアミノ酸残基との間の水素結合によって酵素に固定されていることが示された。 2.コバラミン酵素のコンポジット化合成ペプチド脂質、二分子膜、及び無機高分子マトリックスの3種類のアポ蛋白モデルと、天然型ビタミンB_<12>及び疎水性ビタミンB12類とを組み合わせ、コンポジット生体触媒によるB_<12>酵素類似反応の検討を行った。炭素骨格組み替え反応及び脱ハロゲン化反応などにおいて、従来よりも効率の良い反応系を構築できた。 3.酵素触媒機構の理論化学的解明タンパク質を含まない単純化したモデルを用いて、ジオールデヒドラターゼ反応の各ステップのエネルギーを密度汎関数計算により求めた。反応の必須ステップである水素引き抜きおよび再結合は活性化エネルギーが9.0および15.1kcal/molと、基質の結合エネルギーでまかなえる程度の値であったことから、十分に起こり得る反応であることが示された。
