多発性硬化症における臨床的・免疫学的多様性とその分子遺伝学的背景因子の検討

| Research Abstract |
多発性硬化症(MS)は,複数の遺伝要因と後天的要因との相互作用によって発症に至る,多因子疾患あるいは複合疾患であると考えられている.MSにおける全ゲノムスクリーニングの検索では,HLA領域との連鎖を証明したのみで,この他にも複数の遺伝的因子がMSの発症に寄与すると考えられている.これまで,我々は,vitamin D受容体,estrogen受容体,heat shock protein 70,interleukin-1 β,interleukin-1 receptor antagonist等の遺伝子多型についての検討を行い,その関連性を研究してきた.今年度は,これらに加え,vitamin D binding protein, CD95,β2 adrenergic receptor等のSNPを中心とする遺伝子多型解析を行った.更に,estrogen受容体遺伝子多型と,HLAとの相関なども再検討した.今年度検討した上記3つの遺伝子多型については,患者-コントロール間での有意な相違や,MSの臨床的症状への影響を示唆するデータは得られなかった.遺伝子多型については,人種差によるものも考えられ,今後も,MSの発症及び経過等に深く関与していると考えられるものを中心に,遺伝子多型等をcase-control studyにて解析することにより,日本人におけるMSへの遺伝的要因の関与に迫っていく予定である.
