前立腺癌の再然に関わる機能分子のプロテオミクス解析による同定と診断・治療への応用

Research Abstract	本研究では、前立腺癌におけるホルモン依存性の喪失機序と再燃癌の病態にプロテオミクス解析の手法の応用を試みる。これにより分子標的として再燃前立腺癌の治療の糸口や新しい再燃予知マーカーを得ることを目的とする。今までにアンドロゲン依存性前立腺癌細胞LNCapとアンドロゲン非依存性ヒト前立腺癌細胞株PC-3,DU-145株より、タンパク分画を抽出し、抗チロシンキナーゼ抗体他を用い、ウエスタン法にてそれぞれ機能タンパク質を分離した。それぞれにつき、プロテオミクス解析により更なる分析を進めた。この中で注目すべきチロシンキナーゼ型増殖因子受容体の1つとして、アンギオテンシンII受容体が注目された。 (1)これにつきRT-PCR等でヒト正常前立腺組織および前立腺癌組織、再燃前立腺癌組織の発現パターンを調べ、これら組織にこのアンギオテンシンII受容体が発現していること、また特に前立腺癌および再燃癌で強く発現することを明らかにした。 (2)またこの受容体のブロッカー薬剤より前立腺癌の増殖が抑えられるかどうかを調べ、再燃前立腺癌の増殖抑制に有効なことを明らかとした。またその際のMAPKinaseやSTAT3などの動きを調べ、そのメカニズムを明らかとした(Mol.Cancer Therap.2003)。現在、前立腺組織におけるこの受容体の特異的な型の有無とその発現パターンをプロテオミクス解析により解析中である。
