Outline of Annual Research Achievements

本研究では、複数の脊索動物種を用い、筋肉と骨格の前駆細胞の挙動と胚環境、その背景にある遺伝子相互作用を比較し、筋・骨格・神経系の進化シナリオを復元することを目指す。平成29年度は、体節の一部、皮筋節に由来しながら、遠位方向に長距離を移動する筋芽細胞(migratory muscle precursor; MMP)に注目し、MMPから形成される四肢筋、舌筋、横隔膜の骨格筋の分化制御について解析した。羊膜類のMMPの発生には、Pax3遺伝子、Lbx1遺伝子による筋分化と細胞移動の制御が必要である。円口類ヤツメウナギは四肢(対鰭)を持たないが、舌筋の相同物である鰓下筋を持つ。本年度のヤツメウナギを用いた実験では、遺伝子欠失個体の作成とゲノム配列解析により、Lbx遺伝子が鰓下筋前駆細胞の咽頭部への伸長に必須であることが示唆された。また、硬節および鰓弓骨格のマーカーとしてScxを単離し、その発現領域と筋前駆細胞の位置を比較した。軟骨魚類トラザメを用いた解析では、遺伝子重複により二つのLbx遺伝子が存在し、骨格筋の発生において相互排他的なパターンで発現することが示された。特に鰓下筋においては前方と後方の筋組織(哺乳類のオトガイ舌骨筋、胸骨舌骨筋に相同)がそれぞれ異なる発生遺伝子に制御されて、それらが結合する骨格や神経系との連携を生じながら、発生後期に分化することが示唆された。また、トラザメのLbx遺伝子群は対鰭筋、僧帽筋および体壁筋にも発現し、大きく異なるこれらの筋の分化タイミングに対応する発現パターンを示した。これらの知見から、脊椎動物の初期進化において、皮筋節の腹側が二次的に進展して体幹部腹側を覆う発生機構が獲得され、その吻側部分である頸部筋の複雑化にLbxの発生制御遺伝子の重複が関わったことが示唆された。
