| Outline of Annual Research Achievements |
ω3脂肪酸の健康増進作用において、生体内で生成する機能性代謝物の関与が指摘されている。なかでも、ω3位の水酸化、エポキシ化などの代謝が抗炎症作用などの機能性発現に重要であると考えられている。そこで、このようなω3脂肪酸の代謝に関わる酵素を特定し、その遺伝子欠損マウスを解析することで、代謝経路の生理的意義を明らかにすることを本研究の目的とした。
前年度までの研究で、EPA由来の機能性代謝物17,18-EpETE産生に関わるボトルネック酵素の同定を目指し、ゲノムワイドの脂肪酸酸化酵素スクリーニングを行った。その結果、少なくとも５種類のCYP系酵素が候補に上がった。本年度はそのうちの一つの酵素について遺伝子欠損マウスを用いた解析を行った。このマウスでは、加齢に伴う組織病変が観察された。また、それらの組織から脂質画分を抽出してLC-MS/MSによるリピドミクス解析を行った結果、EPAやDHAのω3エポキシ化代謝物が、野生型と比較して遺伝子欠損マウスで有意に減少していた。このことから、生体内において本酵素はω3脂肪酸の代謝に寄与しており、そこから生成する機能性代謝物がそれぞれの組織における恒常性の維持に寄与していることが示唆された。さらに、最先端のLC-MS/MSシステムを用いた脂肪酸代謝物の包括的リピドミクス解析を用いることで、疾患制御における脂肪酸代謝バランスの重要性について幅広く研究を展開している。
