Outline of Annual Research Achievements

本研究では、磁気マーカーを用いた先端バイオセンシングシステムを開発することを目的としており、本年度に得られた主な成果は以下の通りである。
(1)磁気マーカーの特性解析法の確立。磁気マーカーは様々な環境下で使用され、その動的な磁気特性は環境に大きく依存する。このため、種々の環境下での磁気特性を測定し、マイクロマグネティックスによる数値シミュレーションの結果と比較した。これにより、様々な環境下におけるブラウン緩和とネール緩和の影響を明らかにし、磁気マーカーの磁気特性の定量的評価を可能とした。また、応用に適した特性を持つ磁気粒子のみを選別するための磁気分画法を高度化し、磁気マーカーサンプルを高品質化した。
(2)磁気的免疫検査手法の高度化。洗浄工程が不要な磁気的免疫検査法を高度化するとともに、バイオ物質(C反応性蛋白質:CRP)の検出実験を通して本手法の有用性を示した。すなわち、種々の磁気センサシステムを高感度化してピコ磁気計測を可能にするとともに、磁気緩和法と第3高調波測定法を用いた検査システムを最適化した。また、磁気マーカーの溶液中でのブラウン緩和を利用した検査プロトコールを高度化して本手法の定量性や再現性を高め、洗浄工程なしでの迅速・高感度なバイオ免疫検査を実証した。
(3)磁気イメージング手法の高度化。マルチ検出コイルとFFL(Field Free line)を有する傾斜磁場を組み合わせた磁気粒子の3次元イメージング法を高度化した。本手法は2次元の磁気画像から磁気粒子の3次元濃度分布を再構成できるという他には無い特長を有している。計測システムと画像再構成法を高度化して、磁気粒子の3次元位置と重さに対する検出精度を高めた。また検出実験により、30~50 mm の深部に配置した複数の磁気粒子サンプルの高精度な3次元イメージングを示し、本手法の有用性を実証した。
