| Outline of Annual Research Achievements |
初年度は、IgG4関連硬化性胆管炎（IgG4-SC）と原発性硬化性胆管炎（PSC）の凍結組織から採取したタンパクを用いて、網羅的なプロテオーム解析ならびにリン酸化修飾解析を行った。まず、phosphopeptide enrichment methodを併用したプロテオーム解析で、タンパクのリン酸化修飾が十分検討できることを確認したのち、実際の症例の検討を開始した。最終的に、胆管組織からペプチド23,357個とリン酸化ペプチド4,794個を同定しえた。発現ペプチドに基づいてクラスター解析を行うと、総ペプチドや非リン酸化ペプチドの発現プロファイルはIgG4-SCとPSCで明確な違いはなかったが、リン酸化ペプチド発現プロファイルは両疾患を明確に区別することができ、リン酸化ペプチドが病態の違いをより反映していると考えられた。各疾患で高発現しているタンパクや優位にリン酸化しているタンパクに着目して発現亢進タンパクのネットワーク解析をすると、3つの免疫関連シグナル伝達がPSCに比してIgG4-SCで有意に亢進しており、興味深いことにすべてB細胞や免疫グロブリン関連であった。一方、PSCでは細胞外マトリックスのリモデリングに関連した2つのシグナル伝達が有意に活性化していた。各疾患で過剰発現しているタンパクも同定され、来年度からは個別のタンパクに対して、validation studyを行う予定である。

| Strategy for Future Research Activity |
当初の研究計画に沿って研究を進める。具体的には本年度同定した各疾患で過剰発現しているタンパクに対して、免疫染色などの別の方法を用いたvalidation studyを行う。また、胆道系だけでなく、他臓器のIgG4関連疾患にも応用できるかも含めて検討する。

| Causes of Carryover |
冷却遠心機を購入予定であったが、消耗品の費用が多くかかった関係で、遠心機を購入しなかったことが差額が生じた主な原因である。

来年度からのvalidation studyで使用する消耗品の費用に充てる。
