Outline of Annual Research Achievements

1.本年度はPPIスクリーニング系の構築では、core蛋白質と機能的に相互作用するPA28γのホモダイマーによるmammalian-two hybride screening系の構築をした。化合物スクリーニングLOPAC ライブラリーを使い、数個の候補化合物が確認できた。また、HBVcore promoterアッセイ系を構築し、新規抗ウイルス剤として数種類を同定した。
2.新規培養細胞スクリーニング系によって化合物ライブラリをスクリーニングし、感染および病原性を標的にした化合物同定を行った。プロテオミクス解析から、FKBP6とNS5Aとの相互作用がウイルス複製に重要であることを報告し、FKBP6/8の複合体形成を阻害する化合物同定を行った。
3.CDK阻害剤にHBV複製を阻害する化合物を見つけ、その作用機構を次年度解析する。

Strategy for Future Research Activity

研究計画に変更はなく、同定した抗ウイルス化合物について作用機構解析を行い、新規抗肝炎ウイルス療法開発に繋げる。
