Outline of Annual Research Achievements

本課題では、アレルギー性炎症の病態において、炎症メディエーターとしてのペリオスチンが産生され標的細胞にシグナルを誘導する過程を解明することを目的とする。
1. アレルギー性炎症におけるペリオスチン産生細胞の同定を行うために、ペリオスチンプロモーターの制御下にlacZを発現するノックインマウスを用いて、アトピー性皮膚炎または気管支喘息モデルマウスを作製し、lacZの発現を病理組織学的に観察することでペリオスチンの産生細胞を同定する予定である。
2. ペリオスチンの細胞表面受容体あるいは結合分子の同定を行うために、インテグリンαVβ3を強制発現させたSW480細胞を、ペリオスチンを固相化したプレートに播種し、接着した細胞数を定量するシステムを用い、ペリオスチンにSW480細胞が接着することを確認した。また、この接着はインテグリンαVβ3の中和抗体により阻害されることにより、インテグリンαVβ3を介していることが明らかになった。現在、ペリオスチンの各ドメインのリコンビナントタンパク質を作製・精製中であり、これが完成したら、同じく細胞接着実験によりインテグリンαVβ3の結合部位を同定する。
3. ペリオスチンにより誘導される細胞内シグナル伝達経路の同定を行うために、NIH/3T3細胞にNF-κBのレポーターを導入した。この細胞のペリオスチンまたはインテグリンαVβ3をノックダウンするとレポーター活性が低下し、ペリオスチンによる細胞内シグナルの一部はNF-κB経路を介していることが示唆された。

Strategy for Future Research Activity

インテグリンαVβ3のペリオスチンへの結合配列を同定し、その部分をエピトープとするペリオスチン中和抗体を作製する。また、インテグリンαVβ3以外のペリオスチン結合分子の探索も行う。

Causes of Carryover

実験の進展度合いにより必要物品が予定より少なかったため。
