Outline of Annual Research Achievements

本研究では、ゲノムワイド関連解析（genome-wide association study; GWAS）や全ゲノムシークエンス解析（whole genome sequence analysis; WGS）に代表される、大規模ヒト疾患ゲノム解析および疾患関連遺伝子の情報を、分野横断的なデータベース統合を実施する、大規模インシリコデータ解析を実施する。ドラッグ・リポジショニング対象となる治療薬の効率的なスクリーニングを可能にする新たなゲノム創薬手法の開発が目的となる。平成28年度の研究においては、複数の創薬データベース上に登録された最新の疾患治療薬の網羅的なデータ取得とキュレーションを実施した。1000以上の疾患治療薬標的遺伝子を対象に、治療対象疾患および治療薬とのつながりを網羅した独自のデータベース構築を実施・終了した。特に、治療対象疾患の紐付けが実施できたことにより、各疾患における具体的な治療薬候補の同定が可能となった。さらに、Anatomical Therapeutic Chemical Classification System （ATC分類）に基づき、各種疾患の治療に使用されている治療薬のリストを取得し、上記データベースと紐付けることで、感受性遺伝子情報から、リポジショニング対象治療薬および疾患を自動的にランク付けするシステムの構築を可能にした。国際コンソーシアムとの共同研究において、上記手法の提要を解析中の大規模疾患ゲノム解析データに適用することを提案し、受理されている。適用の過程において、実際に上記システムの有用性が実証され、候補治療薬の新規同定につながるなど、成果が認められている。
