Outline of Annual Research Achievements

昨年度まで卵巣癌を対象とし、スフェロイド培養を行ってきたが、広く癌に共通する抗癌剤耐性メカニズムを検討するため、対象を婦人科悪性腫瘍に拡大した。今年度は、卵巣癌に加え、子宮体癌、子宮頸癌、子宮癌肉腫の臨床検体からスフェロイド培養を実施し、合計卵巣癌５例、子宮体癌４例、子宮頸癌１例、子宮癌肉腫１例のスフェロイド樹立に成功した。スフェロイド細胞を免疫不全マウスの皮下に注入し、腫瘍形成能を確認すると同時に、臨床検体と同様の組織像であることを確認した。樹立されたスフェロイドに対し、Lyoplate screening kitを用いて250種類以上の細胞表面抗原をスクリーニングし、いくつかの癌幹細胞マーカーの同定に成功している。
次に子宮体癌における癌幹細胞の特性を検討するため、幹細胞形質に関与するSex-determining region Y-box 2 (SOX2) 遺伝子に注目し、子宮体癌細胞株を用いてSOX2の機能解析を行った。その結果、SOX2発現は子宮体癌細胞株の細胞増殖に強く関与しており、SOX2がp21のプロモーター領域に結合することで、p21発現を調整し、細胞周期コントロールに影響していることを明らかにした。in-vivo実験系においても、SOX2ノックダウン細胞株では、コントロール群に比し、腫瘍形成が抑制されることを確認した。さらに子宮体部類内膜癌258症例について免疫組織染色でSOX2及びp21発現レベルを調べ、臨床病理学的特徴との関連を検討したところ、SOX2発現は類内膜癌の悪性度及び予後と関連し、SOX2高発現＋p21低発現群では、統計学的に有意に予後不良であり、SOX2とp21が子宮体癌の予後分子マーカーになりうることを明らかにした。
