Research Abstract

脳の性分化メカニズムは未解明の重要課題である。従来の哺乳類の研究から、脳の性分化には生殖腺が合成する性ステロイドが重要な役割を担うと考えられていたが、最近になり、生殖腺に依存しない脳の自立的性分化機構が示唆されるようになった。本研究では、脳の構造と行動に最も典型的な性差を示す鳥類を用い、我々が発見した脳が合成するステロイド(ニューロステロイド)と性決定に関わる遺伝子に着目して、脳の自立的性分化機構の解明を目的としている。本年度は、脳から新規のニューロステロイドを同定した。この新規ニューロステロイドは7α-ヒドロキシプレグネノロンであり、鳥類に限らず脊椎動物の脳に共通して存在する。7α-ヒドロキシプレグネノロンの作用を解析したところ、動物の自発運動を高めることが見いだされた[以上、主論文Proc.Natl.Acad.Sci.USA 101:17282-17287(2004);プレスリリース:読売新聞など]。この新規ニューロステロイド合成には明確な性差があり、雄の脳で多く合成されることがわかった。鳥類の雄は雌に較べて活動性が著しく高いが、これは7α-ヒドロキシプレグネノロンの性差によるもと考えられる[以上、Proc.Natl.Acad.Sci.USA投稿準備中]。7α-ヒドロキシプレグネノロンの性差を導く機構を明らかにするために、このニューロステロイド合成酵素(CYP7b)を脳から同定した。この酵素発現の性差を導く脳内因子としてメラトニンとAd4BP/SF-1に着目して解析を進めた。現在、ニューロステロイド合成の性差を脳の性分化が開始する前後で詳しく解析している。一方、脳の自立的性分化機構の解析のために、鳴鳥の脳から完全長cDNAマイクロアレイを作成した(米国Duke大学と共同)。このマイクロアレイを用い、脳の性分化に関わる遺伝子を同定する計画である。
