| Outline of Annual Research Achievements |
本年度は、環境中の耐性遺伝子の動態解析と評価、遺伝子伝達因子の由来調査および細菌群集解析法の確立を行った。これまでに作業仮説を実験系で断片的に明らかにしてきたが、本年度は魚類を使った大型実験系で耐性遺伝子の消長を明らかにし、養豚場排水・河川・海までの調査で知見を得た。各分担者はレジストーム抽出手法、伝達粒子解析、環境暴露源調査を行なって、本課題の統合的結論へ向けた知見の蓄積ができた。耐性遺伝子生態では、オキシテトラサイクリン（OTC）を投与後のマダイ腸内と飼育環境水での複数のOTC耐性遺伝子の消長を細菌叢変化とともに解析し、腸内細菌叢は比較的安定しているが、多様なtet遺伝子を保有し、一定の遺伝子が定常的に存在しないことが示唆された。また、マクロライド耐性遺伝子と伝達因子の出現を調査した。従来から知られる複数の耐性遺伝子はIntI1との相関があり、IntI1によって環境に残存、拡散してきたことが示唆された。しかし、新規の遺伝子は特定の伝達因子との相関がなく、今後残存と拡散に有利な因子に収束していく可能性が考えられた。メタゲノムでは、環境試料メタゲノムデータからのレジストームデータの抽出方法の確立を目的として、その情報解析基盤を構築した。バイオフィルム解析では、水圏微生物が産生する細胞外膜粒子のDNAは、動物・植物に関与する微生物群由来であることが明らかになった。モデリング・評価では、タイと日本で下水処理場の、耐性菌リザーバとしての評価を行い、下水処理場の活性汚泥中には多くの抗菌薬耐性腸内細菌科細菌が存在することを確認した。

Reason
まだ各項目から統一した結論を得るにはいたっていないものの、各項目は予想以上に進展し、業績も十分得られている。特に、魚類腸内と環境水におけるtetシリーズ遺伝子の動態を明らかにしたことは予想以上の成果である。本研究では、これまでに、遺伝子の環境バックグラウンド形成の解明、二枚貝における耐性遺伝子の分解と蓄積のバランスを解明してきた。本年度は、抗菌剤を投与すると選択圧で耐性菌が増加するのではなく、「養殖魚への薬剤投与の有無に関係なく耐性遺伝子は変遷しながら魚体と環境中に存在する」、という新規の発見が得られた。これらは、これまでに世界的に考えられていた水圏での遺伝子動態の常識を覆す可能性があり、本研究期間内にエポックメーキングができるかもしれない。

多くの新規知見が得られ、論文もコンスタントに発表され、招待講演の件数も多い。また、総説がNature Reviewに掲載されるなど、世界の先端を走る成果がえられてきたので、この成果と発信力をさらに発展させる。また、新規知見から、環境耐性遺伝子のリスクを評価し、将来展望を得ることを目指す。今後は、個々の成果の統合を行なっていく。
