Outline of Annual Research Achievements

目的：環境で薬剤耐性遺伝子が残存・拡散する機構を解明し，リスクになり得るポテンシャルを推定することである。
研究項目と成果：1）耐性遺伝子の残存と伝達の場：未培養菌群集を含む海洋細菌が新規遺伝子sul4のリザーバであることを明らかにした。さらに，バイオフィルムが遺伝子リザーブと伝達の場であることを確証できた。また，遺伝子伝達では，遺伝子供与菌が飢餓状態の時，環境が貧栄養であれば遺伝子伝達はほとんど起こらないが，有機物付加があると伝達能が回復した。
2）環境でのゲノム簡易解析法：スーツケースサイズに収まるゲノム解析セットが開発された。日本とチリの養殖場でテスト中である。
3）リスク評価と軽減策：disability-adjusted life year (DALY)法を応用して耐性菌の健康影響評価法を開発した。さらに日本とタイの環境調査で，都市と病院の排水が耐性遺伝子起源となる高リスク水環境であることを見出した。
意義と重要性：薬剤耐性菌と耐性遺伝子の研究では，人獣臨床のみならず，環境での実態解明が重要である。本研究では，これまで知られていない，海洋細菌の遺伝子リザーバおよび伝達体としての機能を解明し，新たな伝達機構であるベシクル伝達を見出した。さらに，環境現場でゲノム解析が可能な方法の開発および健康影響評価法も開発を行った。このように，本研究では，医療現場以外の自然環境の視点から「ワンヘルス」コンセプトで耐性菌対策を行うための，新規知識と技術を大幅に拡大できた。
