Outline of Annual Research Achievements

近年、これまでの欠点を克服する蛋白質集合体として、蛋白質結晶が注目されている。蛋白質結晶は蛋白質が規則正しく配列した固体状態の蛋白質集合体であり、結晶内部には溶媒チャネルと呼ばれるナノ細孔が形成され、その細孔特性はゼオライトやメソポーラスシリカに匹敵する。また、結晶化により、溶液中の蛋白質や、ランダム会合した状態に比べ、蛋白質の安定性が飛躍的に向上することが知られている。しかしながら、これまで用いてきた蛋白質結晶は、全て単離・精製した蛋白質を結晶化し、架橋化処理を施すことによってはじめてリサイクルや非水溶液中の使用に耐えることができる。また、大量に入手できる安定な蛋白質結晶は限られており、リゾチームを含む数種類の蛋白質の利用にとどまっている。従って、これまでにない新概念の確立が急務となっている。この課題を克服する解決策の一つとして、細胞内蛋白質結晶化反応が挙げられる。昆虫ウイルスが昆虫細胞に感染する際、ウイルスと同時に、結晶性蛋白質である「多角体」が産生され、結晶化の際にウイルス粒子を包埋し、その保護の機能を担うことが知られている。この多角体結晶を機能材料として用いることができれば、結晶化に関わる煩雑な操作が不要となるばかりではなく、遺伝子工学的手法によって、蛋白質をはじめとする様々な分子を内包した機能化蛋白質結晶の作成が可能となる。そこで、本研究では、多角体の細胞内結晶化による多層化機能蛋白質結晶の作成を目指し、(1)細胞内多角体結晶化メカニズムの解明から、(2)精密多層構造を有する多角体結晶の作成、(3)多角体結晶への外来分子の精密多層集積を達成した。
