難治性消化器癌に対する次世代型鉄キレート剤治療の確立

Outline of Annual Research Achievements

肝細胞癌での経口鉄キレート剤の検討：3種類の肝癌細胞株(HepG2, Hep3B, HuH7)に対して鉄キレート剤であるdeferasirox(DFX)の効果MTTアッセイおよびCaspase-3活性で検討し、容量依存的な細胞増殖抑制効果とCaspase-3活性誘導を確認した(in vitro)。さらに肝腫瘍作成マウス（DEN投与により5カ月後肝腫瘍発生）を用いたDFX投与(12週間）においても腫瘍抑制効果を認め、生存率も改善した(in vitro)。さらにDFXによる体重減少もなく、DFXにてALTの有意な低下を認めた。また腫瘍部と非腫瘍部においてHepcidin,Transferrin receptor1,Hif-1αの有意な変動を認めた。
膵癌での経口鉄キレート剤の検討：3種類の膵癌細胞株(BxPC-3, HPAF-II, Panc10.05)において容量依存的な腫瘍増殖抑制効果を認め、低用量(10μM)でS期において細胞周期の休止を、高容量(50, 100μM)でアポトーシスを誘導することを確認した(in vitro)。さらにBxPC-3において塩酸ゲムシタビン(Gem)とDFX併用による相乗効果についても検討したところ、DFX単剤と比較し、有意な増殖抑制効果、Late apoptosisの増加、Caspase活性の上昇を示した。
肝細胞癌では、in vitroおよびin vivoでのDFXの腫瘍抑制効果を確認し、膵癌では、in vitroにおいてDFXの腫瘍増殖抑制効果ならびにGem併用による相乗効果を確認できた。

Reason

肝細胞癌においてはin vitroおよびin vivoの両者から治療効果が確認し、膵癌においてもin vitroの治療効果確認し、おおむね順調である。

Strategy for Future Research Activity

平成29年度は、膵癌においては、膵癌細胞株をヌードマウスに移植したモデルを作成し、DFX単独治療およびGem+DFX併用治療での解析を行う。さらにdeferoxamin (DFO)耐性肝癌細胞株を樹立させ、メタボローム解析を行い、鉄キレート剤の代謝変化を解析し、他の併用治療を探索する。
