| Outline of Annual Research Achievements |
我々は, これまでに, ペリオスチンがマトリセルラータンパク質として機能し, アレルギー疾患において炎症の慢性化を引き起こしていることを示してきた。また, 我々は, ペリオスチンが2型サイトカインであるIL-4やIL-13の下流遺伝子としてSTAT6を介して発現していることを見出したが, その発現機構の詳細は不明であった。このため, 本課題における今年度の研究では, IL-13によるペリオスチン発現機構の解明を目的とした。
肺線維芽細胞をIL-13で刺激した際に誘導される遺伝子をDNAマイクロアレイで網羅的に同定し, その中に転写因子であるSOX11が含まれていた。SOX11の発現をノックダウンするとペリオスチン発現が抑制される一方で, SOX11を強制発現するとIL-13によるペリオスチン発現が上昇した。さらに, IL-13刺激とSOX11ノックダウンを組み合わせてDNAマイクロアレイ解析を行った結果, IL-13誘導遺伝子の中にSOX11依存性と非依存性の遺伝子が存在することを明らかにした。SOX11依存性遺伝子の中にはペリオスチン以外にも炎症や線維化に関連する遺伝子が含まれていた。
以上の結果より, SOX11はSTAT6の下流においてトランスに働く転写因子であるとともに, 肺線維芽細胞におけるIL-13シグナルはSOX11とSTAT6による階層的な転写調節機構により引き起こされ, ペリオスチンもその中に含まれることを明らかにした。
