Outline of Annual Research Achievements

イネのケイ素輸送体OsSIET3とOsSIET5,リン輸送体OsSULTR3;3、鉄輸送体OsOPT7、ムギネ酸輸送体OsVMT、ソバの金属輸送体Nramp5などの発現パターン、組織局在、輸送活性及び変異体の表現型を解析し、それぞれミネラルの輸送に果たす役割を明らかにした。そのうち、OsVMTはイネ節の維管束間の柔組織に発現し、鉄および亜鉛の維管束間輸送過程に関与し、その変異体では結果的に精白米中の鉄および亜鉛の蓄積が1.5~2倍に増加したことを明らかにした。 またイネのカスパリー帯形成に必要な遺伝子OsCASP1を同定し、ミネラルの選択的吸収におけるカスパリー帯の重要性を解明した。さらにリン酸輸送体AtSPDTの解析を通じて、双子葉植物がイネ科植物と異なるミネラルの分配機構を持つことを突き止めた。
導入したLA-ICP-MSを用いて、植物サンプルの元素イメージング法を確立した 。この方法は根、葉、節など様々な植物組織上で、細胞/組織レベルの元素分布を可視化でき、多元素や同位体も同時に検出できる。また定量的イメージング法も開発した。
ミネラル輸送体の結晶構造に関して、ケイ酸輸送体、3種類の金属輸送体、アニオン小分子輸送体の大量発現・精製・結晶化・機能解析を推進した。ケイ酸輸送体はH30年度に解析した立体構造に基づいて機能解析を終了した。また1種類の金属輸送体、アニオン小分子輸送体は低分解能データが得られた。さらにその他2種類の金属輸送体についても精製条件を検討して安定な状態で精製できるようになった。
イネ全体におけるトランスポータ配置の最適性解析を行い、その生物学的な意義を検証した。イネが有害な物質を若い器官に蓄積しないために、どのようなトランスポーター配置を行うべきなのかなどをモデル解析で示す例示研究を行った。

多くのミネラル輸送体の機能解析が予想より早くできた。また数種類のミネラル輸送体の結晶化にも大きな進展が見られた。

Strategy for Future Research Activity

引き続き共同研究者と緊密に連携して、植物ミネラル輸送体の同定、結晶構造解析と輸送モデルの構築を行う。
