Outline of Final Research Achievements

Helper T cells (Th) are called the “control tower” of the immune system, and many subpopulations such as Th1, Th2, Th17 are known in mammals. We already established a method for obtaining clonal Th cell lined from carp. We have already shown that subpopulations exist in fish by examining the characteristics of cloned Th cell lines in carp, but culture of cloned Th cell lines is difficult. Sufficient number of clones was not obtained. In addition, Th cell lines obtained so far were more nonfunctional because they were established from healthy carp. Therefore, in this study, in order to obtain a cloned Th cell line more stably, we prepared a recombinant protein of carp IL-2 and established a method for establishing a stable cloned Th cell line. In addition, we performed mixed lymphocyte reaction of carp and tried to culture alloantigen-specific Th cells.

Academic Significance and Societal Importance of the Research Achievements

近年魚類でもTh亜集団の同定に向けた多くの研究が行われて来ている.しかし,その多くは,魚類Th関連遺伝子の単離と,臓器・組織レベルの発現解析によるものであり,魚類Th亜集団の存在を直接証明しているものは無かった.一方,我々が行ったクローン化T細胞株の作製は,魚類Th細胞を直接証明できる.真骨魚類は,哺乳類と同様,T・B細胞を主体とした獲得免疫機構を持つ.そのため獲得免疫反応の中心的な制御機構を構成するTh細胞の存在や性状を調べることは,魚病の病態解明・予防・ワクチン開発などに大きく貢献する.
