Outline of Annual Research Achievements

LTB4の高親和性受容体BLT1とインバースアゴニストBIIL260の複合体の結晶構造解析に成功した。その結果、BIIL260に含まれるベンズアミジン基が、BLT1のNa+ー水クラスター結合部位に結合していることがわかった。ベンザミジン基内のプロトン化アミジン基は、GPCRで高度に保存されているアスパラギン酸残基と2つのセリン残基にそれぞれ塩橋と水素結合を形成していた。また、ベンズアミジン基内のベンゼン環は、バリン残基とトリプトファン残基と相互作用していた。この相互作用は、Na+ー水クラスターの相互作用を模倣しているため、ベンズアミジン基はNa+ー水クラスターと同様に、不活性状態のBLT1の立体構造を安定化していると考えられた。したがって、ベンズアミジン基はGPCRの7回膜貫通ヘリックス束の活性状態への構造変化を阻害し、Na+ー水クラスターと同様に不活性状態の立体構造の維持に寄与すると考えられた。
また、以前報告されたシステイニルロイコトリエンC4,D4受容体(CysLT2)の恒常活性変異体を参考に、BLT1のヘリックス3のアミノ酸を点変異させたところ、TGFアルファ切断アッセイで恒常活性を示すことを明らかにした。このBLT1変異体を用いたTGFアルファ切断アッセイにおいて、BIIL260は容量依存的にインバースアゴニスト活性を抑制した。本研究によってBIIL260のインバースアゴニスト活性を直接証明することができた。
さらに本年度では、以前同定されたBLT1のインバースアゴニストの候補化合物が恒常活性を抑制するかどうかをTGF切断アッセイを用いて検討した。その結果、BIIL260と同等の活性を有数化合物を複数同定することができた。
