Outline of Final Research Achievements |
Adipose-derived stem cells (ADSCs) were cultured in keratinocyte serum-free medium after co-culturing with fibroblasts on type IV collagen in a medium containing all-trans retinoid acid and bone morphogenetic protein 4. As a result, approximately, 45% of ADSCs were immunostained positively for anti-human cytokeratin 10, and approximately 80% were stained positively for type VII collagen, which is a major component of the anchoring fibrils at the dermoepidermal junction, although undifferentiated ADSCs showed no cytokeratin 10 and type VII collagen immunostaining.

Academic Significance and Societal Importance of the Research Achievements |
脂肪組織由来幹細胞は、細胞作成時に遺伝子操作が不要であることから安全性が高く、抗原性が低く採取が比較的容易であることから、安全性の担保された健常人由来の同種脂肪組織由来幹細胞の臨床応用が期待される。一方、栄養障害型表皮水疱症とは、先天的な遺伝子異常によってVII型コラーゲンが減少または欠損し、表皮・真皮結合部の係留線維に異常を来すことで、全身の表皮が容易にむけて難治な皮膚潰瘍を生じる極めて重症な疾患である。本研究成果は、特定の処理を施した脂肪組織由来幹細胞を、栄養障害型表皮水疱症の治療に応用できる可能性を示した。
