Outline of Annual Research Achievements

我々は組織工学的手法で作製する小口径人工血管の開発を進めている。組織工学的手法としては生体外で細胞培養などの技術を用いて移植片を作成する生体外組織工学や、生体内で移植片を作成する生体内組織工学があるが、いずれの手法も細胞を培養して血管構造を作製したり、細胞にコラーゲンなどの細胞外マトリックスを作製させたりするため、移植片作製に数ヵ月を要し、緊急手術への対応は困難である。そこであらかじめ別の動物体内で移植片を作製・保存することができれば、既製品の人工血管のように緊急手術にも対応可能な血管移植片となりうると仮説を立て、その仮設を動物実験にて検証することを本研究の目的とした。
ビーグル犬皮下にシリコーン円柱基材を1ヵ月間埋入し作製した結合組織管を界面活性剤及びDNA分解酵素を灌流させ脱細胞処理を行った。組織学的評価及びDNA定量にて確実に脱細胞が行われていたことを確認した。また脱細胞前後で引張強度に有意差はなく、処理により結合組織管の強度が損なわれることはなかった。脱細胞処理した結合組織管は1週間冷蔵保存した後にラット腹部大動脈へ異種移植した。移植後の移植片は良好に開存し、1ヵ月後に摘出したところ内腔面は非常に平滑であった。組織学的評価ではグラフト壁内に宿主由来の細胞浸潤が見られ、内腔面には新生内膜の形成が見られた。α-SMA陽性細胞や内皮細胞が層状に形成され移植後早期に血管壁再構築が行われていたことが分かった。
このように生体内組織工学技術に脱細胞処理を加えることで、あらかじめ作製し保存可能な血管移植片として利用できることが期待できる。
