Outline of Annual Research Achievements

我々哺乳類は必須アミノ酸を欠いた食餌を摂取すると摂食抑制を起こすことが知られているが、それが確かに進化の過程で選ばれた9種(Ala, Val, Ile, Thr, Met, Phe, Trp, Leu, Lys)の必須アミノ酸欠失餌だけで起こる現象なのか、あるいはその餌を摂取することで当該アミノ酸の血中濃度が低下するならば(いわゆる)非必須アミノ酸でも同様なことが起こりうるか調べることを目的として実験を行っている。これまでは、数種のアミノ酸を欠いた餌粉末を購入し、それに欠失アミノ酸を補って手練りし、乾燥させたのちに放射線滅菌した餌を実験に使用していたが、手練りの場合、それぞれの餌の形状や粘度が異なり,それによる測定誤差が無視できないレベルにあることが大きな問題となっていた。そこで、高額ではあるが必須アミノ酸1種を欠失したいくつかの固形餌(ペレットタイプ)を購入し、実験に用いることとした。本年度4月に申請者の異動があり、新施設(昭和薬科大学動物実験研究施設)へのマウスの移動とその後の繁殖のため半年近くを費やし、当初の実験計画に遅れを生じたが、それから新たに我々の摂食実験専用の通常とはライトサイクルの異なる飼育部屋を使うことができるようになり、より精密な摂食摂水経過観察実験を行う環境が整った。まず、新規購入したIle欠失、Thr欠失、Val欠失、Met欠失餌で再現実験を試みたところ、今まで通りの実験結果、すなわち必須アミノ酸欠失による早期(最速30分以内)の摂食抑制を確認することができた。

Reason

申請者の異動により、実験に用いるマウスの移動、繁殖維持に半年近くの時間を要したが、その後整った環境下での実験立ち上げにより、より精密な摂食実験を行えるようになった。

Strategy for Future Research Activity

今後は、①摂食抑制の起こるタイミングでの全採血を行い、摂食抑制に先立つ血中濃度の変化を調べ、②Cysが必須となるCys生合成系不全遺伝子改変マウスを用い、通常は非必須アミノ酸であるCys欠失により同マウスで摂食抑制が発生するか調べる。
