Development of disease-modeling technology using hierarchical genome/epigenome editing

Outline of Annual Research Achievements

ゲノム編集については、DNA二本鎖切断(DSB)の修復経路に関わる因子を、ゲノムDNA上の標的領域に集積させることで、MMEJ依存的遺伝子ノックイン(PITCh)をはじめとしたさまざまなゲノム編集を効率化させるlocal accumulation of DSB repair molecules(LoAD)法を確立した。更に、LoAD法を応用し、PITChによる複数遺伝子座への同時ノックインを実現した。これらの成果について、責任著者として論文を取りまとめ、現在国際学術雑誌に投稿中である。
一方エピゲノム編集については、これまでに開発されてきたエフェクター直接融合型やエフェクター集積型の効果を上回る、エフェクター超集積型システム(three-component repurposed technology for enhanced expression; TREE)を新たに開発した。TREEシステムを用いることにより、従来システムでは顕著な発現の活性化がみられなかったがん関連遺伝子の発現を強く誘導できることが明らかとなった。更に、TREEシステムの優位性が細胞種や遺伝子座に依存しないことを証明し、普遍的に高い効果を示すエフェクター集積システムとして利用可能であることが示唆された。本成果についても、責任著者として論文を取りまとめ、現在国際学術雑誌に投稿中である。また、「標的遺伝子にエフェクタータンパク質を集積するための組成物、およびその利用」として特許出願を完了した(特願2018-041322)。
これらの成果により、ゲノム編集・エピゲノム編集の両面から、多因子性の複雑な疾患の発症をモデリング可能な技術が確立された。
