Outline of Annual Research Achievements

多能性幹細胞は、naive型多能性幹細胞(NSC)とprimed型多能性幹細胞に分類される。ヒトES/iPS細胞は、分化する能力が限定されており、NSCよりも僅かに分化しているEpiblast stem cells (EpiSC)と定義される。naive型は着床前の内部細胞塊でみられる多能性に近い状態であり、Primed型は着床後のEpiblast内でみられる多能性に近い状態である。
人工的組織幹細胞であるiTS細胞は、前駆細胞や幹細胞などの中間状態を持つ部分的にリプログラムされた細胞である。通常、iTS細胞は体細胞を山中因子でリプログラミングしてから約10日後に取得できるが、理論上、iPS細胞から分化誘導することにより樹立が可能と考えられる。
iPS細胞は、膵臓系統に分化させるin vitroプロトコールはいくつか報告されているが、これらのiPS細胞からのβ細胞生成に関するデータは限られている。そこで、我々は NSCにおける増強された分化万能性に着目し、NSCからiTS細胞を経由した分化誘導法により膵臓β細胞へ効率的に分化できるかどうかを検証することを本研究の目的とした。
乳歯歯髄細胞由来のiPS細胞をNSCへ転換し、胚様体(EB)を形成した。その胚様体を、β細胞系統へin vitroで分化誘導することで、β細胞特異的マーカーを発現し、コロニー状を呈する膵iTS細胞の樹立に成功した。逆に、EpiSC由来のEBから分化誘導では、in vitroでそのようなコロニー形成を示さなかった。ヌードマウスへの膵iTS細胞の膵臓内移植では、顕著な腫瘍形成なしに、ヒト由来のPDX1陽性インスリン産生細胞を含む細胞塊を生成した。これらのNSC-iTSC分化誘導系は、1型糖尿病を治療するためだけでなく、他の組織由来細胞への分化誘導においても有望なリソースとして使用できると考えられた。
