Outline of Annual Research Achievements

本研究課題では、糞便、唾液、血液といった体への負担が少なく頻回検査が可能なサンプルを用い、腸内細菌叢の16Sメタゲノム解析による、大腸癌の新たな診断法の確立を目指し研究を行ってきたが、遺伝子解析を担当する共同研究者の死亡により腸内細菌叢の遺伝子解析が困難となった。遺伝子解析体制の再構築を試みたがコロナウイルスの再流行などあり、困難であり、腸内細菌に関連した研究の続行は不可能であった。そこで、検討するマーカーの対象を広げ、脂質、腫瘍の遺伝子解析や炎症性マーカーを用いた予後分析、抗癌剤の効果への影響の分析を続行した。今年度は解析と論文化にとどめ、新規の測定などは施行しておらず、研究費は使用しなかった。
大腸癌のKras遺伝子変異、Circulating DNAが大腸癌の予後不良因子であることを明らかにし、データベース解析により、肝転移予後予測に有用なノモグラムを作成した。
遺伝子変異の予後への影響（Prognostic Value of KRAS Exon-specific Mutations in Patients With Colorectal Cancer, Asako K et al. Anticancer research）、リキッドバイオプシーによる予後予測（Analysis of Circulating DNA to Assess Prognoses for Metastatic Colorectal Cancer Patients Treated with Regorafenib Dose-Escalation Therapy: A Retrospective, Exploratory Analysis of the RECC Trial, Ohta R et al. Digestion)等を論文化した。
