Outline of Final Research Achievements

Synovial mesenchymal stem cells from rheumatoid arthritis patients osteoarthritis patients had similar proliferation property, colony formation property, differentiation capacity, and surface antigen profiles. In suspended synovial tissue culture model, synovial mesenchymal stem cells from rheumatoid arthritis patients and osteoarthritis patients had similar average cell yield, cartilage differentiation property and surface antigen profile, though the cell yield variation was larger among rheumatoid arthritis than osteoarthritis. Both results were reviewed and accepted in the international scientific journal.

Academic Significance and Societal Importance of the Research Achievements

変形性膝関節症および関節リウマチ症例の滑膜幹細胞の増殖能・分化能の違いを明らかにすることが、関節リウマチにおける滑膜炎による関節破壊の病態生理の解明や滑膜幹細胞による再生医療の安全性と治療効果を増幅させる手法の開発に有用である。また関節リウマチ患者に滑膜幹細胞治療を応用する道を開くだけでなく、既存の対象患者に対する移植医療においても培養滑膜幹細胞の品質評価や細胞収量および分化能・移植後の拒絶・炎症反応等、安全性の問題の解決につながる。さらに、幹細胞の増殖・分化を促すサイトカインの解明は、炎症時の滑膜間葉系幹細胞の挙動解明の助力となり、さらには関節内組織損傷における自然治癒過程の解明にも役立つ。
