Outline of Annual Research Achievements

本研究では、高脂肪食をKKマウスに8週間負荷し糖尿病性腎症モデルを作成した。この高脂肪食負荷KKマウスの腎臓では、グロボ系糖脂質Gb3の発現が著しく増加していた。その分子種解析を, 液体クロマトグラフ-タンデム質量分析計(LC-MS/MS)を用いて行った結果、Gb3のセラミド部分のアシル鎖に飽和の極長鎖(C22, C23, C24)有する分子種が特に増加していることを見出した。この結果より, グロボ系糖脂質の病態形成への関与が示唆されることから, 糖尿病性腎症の病態形成に重要な炎症反応における, グロボ系糖脂質の炎症惹起活性の有無をヒト単球やマウス骨髄由来マクロファージを用いて評価した. その結果, 極長鎖グロボ系糖脂質は, Toll-like receptor 4 (TLR4)リガンド(LPS, HMGB1)の存在下, TLR4選択的なポジティブモジュレーターとして炎症促進活性を持つことが示唆された。また、彼はレプチン受容体が変異したdb/dbマウスではGb3の発現が低く、レプチンの腹腔内投与で、Gb3の発現が回復することを見いだしている。高脂肪食負荷により肥大した脂肪組織から分泌促進されたレプチンが、腎臓のグロボ系糖脂質の発現を増加させ, 増加したグロボ系糖脂質が、TLR4に作用することで炎症反応を促進し, 糖尿病性腎症の病態形成に関与している可能性が本研究より示唆された。これらの研究成果は、糖尿病性腎症の新たな診断法や治療薬開発につながることが期待される.
