Outline of Annual Research Achievements

本研究では、低濃度・中濃度・高濃度の鉛濃度が検出された子供を対象に、血中・尿中・糞便中の鉛濃度の推移および糞便を用いた腸内細菌叢解析を行い、鉛暴露が腸内細菌叢に及ぼす影響を解明する。
本事業中にザンビアに5回渡航しカウンターパートのJohn Yabe講師とディスカッションおよびサンプリングを実施した。現地で必要となるゲノム抽出装置、PCR装置などのラボのセッティングは初年度前半の渡航で完了した。約800名の子供から血液、尿、および糞便を収集した。現地クリニックにおけるオンサイト血液中鉛濃度測定を終了し、低濃度から高濃度の広範囲における血中鉛濃度を示す検体を集めた。糞便由来の腸内細菌叢解析を実施するためザンビア大学獣医学部に設置したラボにおいて細菌ゲノムを抽出した。これらの試料を対象に金属濃度および細菌叢解析を実施し、大規模なデータ取得が完了した。現在、これらのビッグデータを解析している段階である。対象を人だけでなく、人と生活圏を共有する野生ラットにも拡大し、血液、各種臓器、胃内容物、糞便を採取し、人では不可能な臓器中の金属濃度と細菌叢との関連を明らかにする。
共同研究の重要な目的である途上国における環境学・毒性学の研究者育成のためにザンビア大学より研究者を招聘し技術移転を実施した。招聘者はザンビア大学の学生に教育・指導を実施しており、当該分野の推進に貢献している。
2019年度より治療プロジェクトを遂行する世界銀行(World Bank)の担当者と実施計画のディスカッションを実施した。本共同研究のカウンターパートであるJohn YabeはWorld Bankプロジェクトにおけるメンバーでもあり、当該研究との連携の役割を担うことを確認した。

Reason

昨年度に引き続きザンビアに渡航し共同研究を推進した。当該年度も試料採取を実施した。これらの試料を対象に当該年度も金属濃度および細菌叢解析を実施し、大規模なデータ取得が完了した。現在、これらのビッグデータを解析している段階である。さらに本年度は対象を人だけでなく、人と生活圏を共有する野生ラットにも拡大し、血液、各種臓器、胃内容物、糞便を採取した。これらの試料は人では不可能な臓器中の金属濃度や細菌叢との関連を明らかにすることができる優位性を持っている。
共同研究の重要な目的である途上国における環境学・毒性学の研究者育成のためにザンビア大学より研究者を招聘し技術移転を実施した。招聘者はザンビア大学の学生に教育・指導を実施しており、当該分野の推進に貢献している。
鉛濃度の高い住民に対する治療プロジェクトを遂行する世界銀行(World Bank)の担当者であるPaul MukukaおよびRuma Tavorathと実施計画のディスカッションを実施した。本共同研究のカウンターパートであるJohn YabeはWorld Bankプロジェクトにおけるメンバーでもあり、当該研究との連携の役割を担うことを確認した。

Strategy for Future Research Activity

最終年度となる３年度目については、これまでにデータを取得済みの金属濃度およびシークエンス解析のデータにおける関連性を多変量解析などの統計学的解析をもとに明らかにする。得られたデータをもとにマウス・ラットなどの実験動物を用いた暴露実験からデータの再現性を実証し、金属暴露が腸内細菌叢に及ぼす影響を解明する。新型コロナウイルス感染症の拡大により、予定通りにザンビアへの渡航ができない可能性もあるが、既に充分な試料は入手しており、解析データも取得していることから支障はないと考えている。
