延髄孤束核へ神経型一酸化窒素合成酵素のタンパク質の直接導入による循環機能への影響

Research Abstract

延髄孤束核は、動脈圧受容器よりの脳内terminalであると知られている。そして、一酸化窒素(nitric oxide,NO)は孤束核で循環調節に関する神経伝達に対し興奮性に作用することが明らかになっている。さらに、孤束核へ一酸化窒素合成酵素(NO synthase、NOS)の遺伝子導入で血圧変化を調べ、脳へ遺伝子を導入することによる遺伝子治療の可能性が検討されている。しかしながら、遺伝子導入では慢性実験が必要であり、体内でゲノム情報から作られるタンパク質の機能をすぐに解析することはできない。私達は、in vivoで脳内の特定機能部位へβ-galのタンパク質を直接導入することに既に成功している(Neuroscience Letters、378:18-21、2005)。この方法により、そのタンパク質の機能を急性的に解析することができる。平成18年度は神経型NOS(neuronal NOS、nNOS)のタンパク質をvectorとともに、孤束核へ微量注入し、血圧・心拍数・平均血圧の変化を3時間にわたって観察し、平均血圧はβ-galとvectorのcontrol群と比べ1時間後より有意に下降し、注入されたnNOSのタンパク質が機能を果たしていることを確認じた。平成19年度は、12時間後も平均血圧はcontrol群と比べ有意に下降することを調べた。また、タンパク質の導入部位は孤束核であることも確認した。これらより、脳の特定機能核へ導入したタンパク質は機能を発揮していることが判明した。今後、タンパク質治療という新しい分野を開拓できるかも知れない。
