縫合閉鎖から読み解く頭部骨数の減少進化

申請者らの近年の研究によって脊椎動物の頭蓋の相同性についての従来理論は全面的な再検討を要することがわかってきたことを踏まえ,骨間の縫合閉鎖という現象に注目した比較形態学的研究を進め,縫合閉鎖過程の多様性と共通性,その進化的背景に関する研究を進めた.30目78科189種の3000標本の胚標本のCT撮像を行い,骨CT撮影およびヨウ素コントラスト増幅法を用いた液浸標本撮影を行った.これらにより,骨だけでなく神経,血管,筋,軟骨の再構築を行い,これらの比較解剖学的な観察記載を進めた.また,トカゲ,カメ, ニワトリをはじめとするの頭部組織切片を作成し,免疫染色とヘマトキシリン=エオシン染色を行った.これらの動物の組織切片をAmiraで三次元再構築し,胚における脳・神経・骨格・筋・血管の精確な三次元形態モデルを作成し,各パーツの立体的な相互配置,発生に伴う骨成長,骨の縫合閉鎖パターンを把握した.分析の結果,Dlx5とLmx1bは脳の拡大と関連しているとともに,これらが頭蓋骨の縫合閉鎖を促進するがわかった.脳の発達に不可欠なシグナリングは同時に,頭蓋骨の成長に大いに影響を与えることが示唆された.相対的な脳のサイズの増加が単弓類の進化の重要な側面の一つであるということを考えると、頭蓋骨の単純化もまた脳のサイズの進化と関連している可能性がある.仮説的なシナリオとして,大脳化がDlx5やLmx1bといった遺伝子が選択されたが、これらの遺伝子は頭蓋骨のさまざまな領域で早期の縫合閉鎖と骨間の癒合の促進を引き起こしたと考えられた.加えて,頭蓋の縫合を開放状態に保つTwist1が,大脳化が特に進んだ哺乳類で選択された可能性が示唆された.
