ロドプシンの多彩な機能をもたらす構造ダイナミクスの解明

Outline of Annual Research Achievements

我々は2013年以降、それまでに予想もされていなかった微生物由来の新規ロドプシンの存在を次々に報告し、光駆動ナトリウムポンプについては立体構造も決定した。その結果、微生物ロドプシンは共通の構造と光反応を持つにも関わらず、実に多彩な機能を示すことがわかってきた。本研究では、構造と機能をつなぐタンパク質の構造ダイナミクスを明らかにすることで、共通構造から多彩な機能が生じる機構を解明することを計画した。研究では、我々が世界をリードする赤外差スペクトル分光法を中心として、光反応中間体の構造解析を行う。低温法、時間分解法、全反射法などの手法を駆使して光反応中間体におけるレチナールやタンパク質部分、内部結合水の構造変化を明らかにすることで、種々のロドプシンにおけるメカニズムを明らかにすることを目指した。
光駆動ナトリウムポンプKR2の構造ダイナミクスについては、Q123変異体の低温赤外分光法により細胞質側に存在する水素結合ネットワークの変化を明らかにした。また、時間分解赤外分光法を適用することで、O中間体が13シス型のレチナール構造を持つことがわかった。さらにKR2の全反射赤外分光法により、ナトリウムイオンの結合によりチロシンがきわめて強い水素結合を形成することがわかった。
内向きプロトンポンプPoXeRの構造ダイナミクスについては、13シス型から2つの異性化反応の経路があることを明らかにした。また、陰イオンチャネルロドプシンの研究においては、米国との国際共同研究によりNature誌に2報の論文を発表することができた。酵素ロドプシンRh-PDEについては初めて高精度赤外分光計測を発表した。さらに初年度には、イスラエルとの国際共同研究により、既知の微生物ロドプシンと配列の相同性を持たないロドプシンを発見し、Nature誌に発表した。

Reason

1年で3報の論文をNature誌に発表したという事実は、今後、再現不可能なほどの成果であり、きわめて顕著なものである。当初、計画した光駆動ナトリウムポンプ、内向きプロトンポンプ、チャネルロドプシン、酵素ロドプシンのすべてにおいて初年度から論文発表が実現しており、質量ともに世界をリードするに相応しい成果である。光駆動ナトリウムポンプについては、超高速分光、共鳴ラマン散乱、固体NMR法、高速AFMを用いた共同研究による新たな知見も論文発表することができた。
さらに、我々がヘリオロドプシンと名付けた新規ロドプシンは、既知のロドプシンと配列相同性がないにも関わらずよく似た構造ダイナミクスを示すことが明らかになっており、さらなる大きな発展性を示している。

Strategy for Future Research Activity

当初の予定通りの研究を推進する。一方、2018年に新たに発見したヘリオロドプシンの研究も大いに楽しみである。
