Outline of Annual Research Achievements |
A new scheme is designed to achieve the objectives based on voltammetric approach. For this, sequences of two peptides are optimized and synthesized. 1. CatE binding peptide aptamer 2. CatE substrate peptide Next, in order to integrate both the peptides onto the surface of voltammetric sensor in functionally active form, a linker peptide chain composed of alpha-strand property (18 amino acids) and PEG-2000 is designed and evaluated. As a result, detection limit of model target (amyloid beta42) could be increase as low as 4 nM using voltammetric DEPSOR sensor and disposable screen printed electrochemical chips.

2: Research has progressed on the whole more than it was originally planned.
Reason
In order to make a simple POCT sensor (demand from one of the co-investigator in this project from medical-side), an alternative plan by utilization of simple sensing surface of voltammetric approach is designed and that's bit delayed the scheduled experimental plan.

Strategy for Future Research Activity |
Both the peptides (#1 and #2) with alpha-strand chain will be custom synthesized and coupled with nanoparticles for electrochemical analysis using DEPSOR. In the first stage, a recombinant Cathepsin E will be measured and limit of detection will be measured. After achieving a desired sensitivity, the scheme will be demonstrated for clinical sample obtained by one of the co-investigator from hospital.

Causes of Carryover |
The budget will be utilized to purchase the reagents and to hire a researcher to perform voltammetric experiment. Some amount will be used for travel too.
