Outline of Annual Research Achievements |
A silicon-based FET sensor is developed to measure expression-level of Cathepsin E (CatE). This was achieved by immobilizing the CatE specific substrate peptide onto the surface of FET followed by measuring the cleavage of CatE using a palm-sized portable FET device (termed FETSOR). The measurement conditions for the working of biosensor were optimized at pH 7.4 and 0.01X PBS and we could successfully differentiate between the presence and absence of 2.3 pmol CatE’s activity to cleave the substrate peptide. The overall charge on the peptide at pH 7.4 is +8.51 and after the cleavage it reduces to +3.61 which was confirmed by decrease in the gate voltage measurement by 0.05V. Therefore, we successfully demonstrated and evaluated the utility of FETSOR for the measurement of CatE.

We are continuously searching a collaborator for providing clinical samples after the pre-retirement of Prof Yamashita from Hokkaido University.

Strategy for Future Research Activity |
First, we will find a collaborator in some hospital for providing the clinical samples. Then, we will arrange to install the FETSOR system in the hospital for clinical sample testing and evaluation.

Budget will be utilized to conduct the clinical sample testing and evaluation.
