Outline of Annual Research Achievements |
A silicon-based FET sensor is developed to measure expression-level of Cathepsin E (CatE). This was achieved by immobilizing the CatE specific substrate peptide onto the surface of FET followed by measuring the cleavage of CatE using a palm-sized portable FET device (termed FETSOR). The measurement conditions for the working of biosensor were optimized at pH 7.4 and 0.01X PBS and we could successfully differentiate between the presence and absence of 2.3 pmol CatE’s activity to cleave the substrate peptide. The overall charge on the peptide at pH 7.4 is +8.51 and after the cleavage it reduces to +3.61 which was confirmed by decrease in the gate voltage measurement by 0.05V. Therefore, we successfully demonstrated and evaluated the utility of FETSOR for the measurement of CatE.

Remarks |
We conclude the project with the achievement of developing a portable peptide-based FET sensor for a simple and rapid detection of breast cancer biomarker Cathepsin E.
