Role of small GTPase signaling in the direct pathway of basal ganglia

Outline of Annual Research Achievements

本研究では、背側線条体に存在するドーパミンD1受容体陽性細胞 (D1R+細胞) から黒質網様部へ投射する直接路が司る運動制御の分子メカニズムを解析した。側坐核のD1R+細胞において、protein kinase A (PKA)-small GTPase Rap1シグナル伝達系が、報酬行動に重要な役割を果たすことが知られているので、まず、上流で機能するPKAに注目した。独自開発したダブルAAVベクターシステムを利用して、直接路特異的にPKAを活性化したマウスを作製した。最初に、頭部片側のD1R+細胞でPKAが活性化されたマウスを用いてtail hanging testを行なったところ、PKAシグナリングが活性されている側とは反対の方向に体を曲げるという特徴的な行動が認められた。次に、頭部両側のD1R+細胞でPKAが活性化されたマウス (PKA-CAマウス) を用いてopen field testを行なった結果、コントロールマウスと比較して自発運動量の有為な減少が認められた。また、メタンフェタミン投与後の運動量も有為に減少した。運動量減少の原因を解明するために、PKA-CAマウスのステレオ行動を解析中である。PKA-CAマウスの黒質網様部における自発発火を記録したところ、自発発火頻度、自発発火パターンには影響が認められなかった。大脳皮質から線条体への入力を模倣するように、大脳皮質運動野を電気刺激して黒質網様部で応答を記録したところ、コントロールマウスと同様に「早い興奮-抑制-遅い興奮」という三相性の応答が認められたが、抑制の持続時間が長くなっていた。抑制は、直接路を介する応答であるため、PKAの活性化により、直接路の神経活動が上昇していると考えられる。以上の結果から、背側線条体のD1R+細胞におけるPKAシグナリングは、直接路による運動制御に重要な役割を果たすことが示唆された。
